She was born on July 30, 1979, at Troyes. Aube to civil servants parents. She has two elder sisters Annelise  Hesme, born on May 11, 1976,  and Elodie Hesme, born on May 10, 1973

Like Clotilde, both her sisters are also famous actresses working primarily in French cinema. 

While completing her study on drama from the prestigious Cours Florent, she acted on various plays. 

She also was a member of the National Conservatory of Dramatic Art for more than ten years.

Her first lead role was in Philippe Garrel's Regular Lovers. She has won the SACD Award for Female Revelation and Caesar nomination for the most promising actor for Christopher Honore's Love Songs.

After winning Caesar Award for most promising actress in 2011 for the French Film Angele and Tony directed by Alix Delaporte, she came into popularity. 

Les Revenants (2012) is her internationally acclaimed and successful work. The actress is also part of the popular Netflix series Lupin (2021).
